The study of hereditary retinoblastoma led to the development of tumorigenesis that might be induced by two mutations, ultimately resulting in the cloning of the RB1 gene. Since then, the two-hit theory has aided in the identification of several tumor suppressor genes (TSG). Recently, descriptive analyses have been conducted on the functions of haploinsufficiency, gene dosage consequences, and epigenetic control in a few TSGs, including PTEN, P16, and P53.
